Association between asporin gene aspartic acid repeat polymorphism and osteoarthritis: an updated meta-analysis

چکیده
Objective: Osteoarthritis (OA) is a common disease characterized by progressive degeneration of articular cartilage. Several studies have been performed to evaluate the association between asporin gene aspartic acid repeat (ASPN D-Repeat) polymorphism and osteoarthritis susceptibility, but produced conflicting results. Our aim is to determine whether ASPN D-repeat polymorphism is associated with susceptibility to OA. Methods: A comprehensive literature search for all relevant studies was performed. The association between ASPN D-Repeat polymorphism and OA in each ethnic group was investigated. Sensitivity analysis and publication bias test were conducted. Results: Eleven studies with 8463 participants (4842 OA patients and 3621 controls) were enrolled in this study. The combined results revealed no significant association between D13 allele and the risk of OA (D13 allele vs. other alleles combined: OR 0.939, 95% CI 0.844-1.045). Subgroup analysis stratified by ethnicity and OA site further confirmed the irrelevance. The pooled results on the associations between other alleles (D14, D15, D16 and D17, respectively) and the risk of OA were similar to those of D13 allele, no significant association was identified in the overall population or in each subgroup population. Conclusion: This meta-analysis suggested that ASPN D-repeat polymorphism is not associated with OA susceptibility. Since potential confounders could not be ruled out completely, further well-designed studies are required.

An updated meta-analysis of the asporin gene D-repeat in knee osteoarthritis: effects of gender and ethnicity
BACKGROUND Knee osteoarthritis (KOA) is the most prevalent form of knee joint disease and characterized by the progressive degeneration of articular cartilage. Although pathology of KOA remains unknown, genetic factors are considered to be the major cause. Asporin is a group of biologically active components of extracellular matrix (ECM) in articular cartilage, and asporin gene (ASPN) D-repeat ...

Osteoarthritis (OA) is the most common form of human arthritis. Genetic factors have been implicated in OA. It was reported that an aspartic acid (D)-repeat polymorphism in the gene encoding asporin (ASPN) was associated with OA of knee and hip joints in Japanese; in the three independent studies performed, the D14 allele of the ASPN polymorphism was over-represented and the D13 allele was unde...

Association of the D repeat polymorphism in the ASPN gene with developmental dysplasia of the hip: a case-control study in Han Chinese
INTRODUCTION Developmental dysplasia of the hip (DDH) is a common skeletal disease, which is characterized by abnormal seating of the femoral head in the acetabulum. Genetic factors play a considerable role in the etiology of DDH. Asporin (ASPN) is an ECM protein which can bind to TGF-β1 and sequentially inhibit TGF-β/Smad signaling. A functional aspartic acid (D) repeat polymorphism of ASPN wa...
متن کاملLack of association of a variable number of aspartic acid residues in the asporin gene with osteoarthritis susceptibility: case-control studies in Spanish Caucasians
A recent genetic association study has identified a microsatellite in the coding sequence of the asporin gene as a susceptibility factor for osteoarthritis (OA). Alleles of this microsatellite determine the variable number of aspartic acid residues in the amino-terminal end of the asporin protein. Asporin binds directly to the growth factor transforming growth factor beta and inhibits its anabo...
